1. INTRODUCTION - INTRODUCTION
L’ob je t d e l’é tud e é tait d ’é value r le p ote ntie l

in vitro. Une ap p roche e xp é rime ntale b asé e sur la
me sure d u taux d e mé lanine e t d e la coloration
d e s é p id e rme s ap rè s traite me nt top iq ue a é té
proposée.
The p urp ose of the stud y was to asse ss the
white ning p ote ntial of a cosm e tic finishe d p rod uct
using reconstructed human tanned epidermis. An in
vitro e xp e rim e ntal ap p roach b ase d on the
m e asure m e nt of m e lanin conte nt, and coloration of
tissue s afte r top ical tre atm e nt with the te st p rod uct,
was proposed.

traite me nt d e s tissus. Ap rè s incub ation, le milieu
d e croissance e st é liminé e t re mp lacé p ar d u
milie u ne uf. Le p rod uit à l’é tud e e st ap p liq ué , à
raison d e 5 mg / cm 2, d ire cte me nt à la surface d e s
épidermes : 2.5 µl d e p rod uit sont d é p osé s à l’aid e
d ’une microp ip e tte e t é talé s, sur la surface d u
stratum corne um, à l’aid e d ’un ap p licate ur. Le s
tissus sont e nsuite re mis à incub e r à 37°C, e n
é tuve air-CO2. La formule -te st e st ap p liq ué e à J0,
J1, J2, J3, J6 et à J7.
A la fin d u traite me nt (J8), le s p aramè tre s (taux d e
mélanine et coloration des tissus) sont analysés.
Coloration des épidermes
La coule ur d e s é p id e rme s e st me suré e à J0 (avant
ap p lication d e s p rod uits) e t 24h ap rè s la d e rniè re
ap p lication, à l’aid e d ’un chromamè tre Minolta CR
200. Ce t ap p are il conve rtit le s coule urs situé e s
d ans la p lag e d e p e rce p tion humaine e n un cod e
numé riq ue comp osé d e trois p aramè tre s : L* q ui
ind iq ue la clarté e t a * e t b * q ui sont le s
coord onné e s d e chromaticité . a* e t b * ind iq ue nt
le s se ns d e s coule urs : + a * va ve rs le roug e , -a *
ve rs le ve rt, + b * va ve rs le jaune e t -b * ve rs le
b le u. Le s p aramè tre s L*, a* e t b * sont me suré s sur
chaq ue é p id e rme s. Le s me sure s sont ré alisé e s e n
trip licate e t le s vale urs moye nne s d e chaq ue
p aramè tre sont calculé e s. L’ang le Typ olog iq ue
Ind ivid ue l (ITA°) q ui d é finit le d e g ré d e
p ig me ntation d e la p e au (p lus l’ITA° e st faib le ,
p lus la p e au e st p ig me nté e ) a é té calculé se lon la
formule : ITA° = [Arc tan (L* - 50)/b*] x 180/
Taux de Mélanine
A la fin d e l’e ssai, le s é p id e rme s sont d é taché s d e
le ur sup p ort à l’aid e d ’un scalp e l. Le s tissus sont
mis à chauffe r p e nd ant 45-60 minute s à 100°C,
d ans d u SOLVABLE™. Ap rè s re froid isse me nt d e s
e xtr a its e t ce ntr ifug a tion, d e s a liq uots (100µl) sont
prélevés e t déposés d ans une p laq ue 96 p uits.
L’absorbance des solutions est lue à 405nm à
l’aide d’un lecteur de microplaque. Parallèlement,
une gamme étalon construite à l’aide de mélanine
synthétique (Sigma) et mise à incuber à 100°C
pendant le même laps de temps que les extraits
cellulaires, permet de transformer les densités
optiques (D.O405nm) en Mélanine (µg/ml).

for 3-4 hours, in the incub ator at 37°C in a
hum id ifie d atm osp he re with 5% CO 2 p rior to
te sting . Afte r incub ation, the m e d ium und e rne ath
tissue s was re p lace d b y fre sh m e d ium The p rod uct
to b e te ste d was ap p lie d top ically at 5 m g / cm 2 onto
the e p id e rm is, using an autom atic p ip e tte : 2.5 µl of
te ste d p rod uct we re d e p osite d with a m icrop ip ette
and sp re ad onto the surface of stratum corne um
using an ap p licator. The tissue s we re conse q ue ntly
incub ate d , at 37°C with a 5% CO 2 atm osp he re . The
te st p rod uct was ap p lie d d aily from Day 0 to Day 3
and at day 6 and day 7 (i.e 6 applications).
At the e nd of the tre atm e nt p e riod (Day 8), skin
coloration and melanin content were analyzed.
Epidermis coloration
Ep id e rm is color m e asure m e nt is p e rform e d b e fore
starting tre atm e nt (Day 0) and 24 hours afte r the
last ap p lication of the te st p rod uct (Day 8). The
coloration is q uantifie d with a chrom am e te r CR-200
Minolta, which conve rts colors locate d within the
rang e p e rce ive d b y hum ans to a d ig itize d cod e
com p ose d of thre e p aram e te rs L* for re lative
lig htne ss (from d ark to p ale ), a* for the b alance
b e twe e n re d ( +a*) and g re e n ( - a*), and b * for the
b alance b e twe e n ye llow ( +b *) and b lue (-b *). The
L*, a* and b* values of the three-dimensional space
d e fine d b y the CIE are re g iste re d on e ach
e p id e rm is. The m e asure m e nts are p e rform e d in
trip licate , and the m e an value s of L*, a* and b * are
calculate d . The Ind ivid ual Typ olog y Ang le (ITA°)
which d e fine s the d e g re e of skin p ig m e ntation
(d arke r skins have lowe r ITA°) is calculate d as
follows: ITA° = [Arc tan (L* - 50)/b*] x 180/
Melanin content
For e ach te ste d p rod uct or controls, at the e nd of
the te st p e riod , the culture s we re re m ove d from the
p olycarb onate inse rt b y cutting out the filte r on
which the tissue s we re g rown, with a sharp scalp e l.
Each filte r was p lung e d into SOLVABLE™ and he ate d
at 100 d e g re e s Ce lsius for 45-60 m inute s. Afte r
cooling and ce ntrifug ation, aliq uots (100 µl) we re
ad d e d into 96-we ll m icrop late . Op tical d e nsity
(O.D405) is re cord e d at 405 nm with a m icrop late
re ad e r. Concurre ntly, synthe tic m e lanin stand ard s
(Sig m a) are incub ate d in the sam e cond itions. This
stand ard curve allows the transform ation of the
O.D405nm into Melanin Unit (µg/ml).

3. RESULTATS - RESULTS
3.1 ACCEPTABILITE CUTANEE DU PRODUIT - CUTANEOUS ACCEPTABILITY OF PRODUCT
L’e ssai a é té ré alisé sur d e s é p id e rme s EPISKIN
non p ig me nté s fournis p ar SKINETHIC™ (Lot N° 09EPI-001, surface 1.07 cm 2) afin d ’é value r
l’acceptabilité cutanée du produit [08.922].
La viab ilité d e s tissus a é té é valué e p ar un te st
MTT.
The assay was performed on un-pigmented EPISKIN
e p id e rm is (1.07 cm 2 tissue s, Batch n° 09-EPI-001)
sup p lie d b y SKINETHIC™ to e valuate the cutane ous
acceptability of the product [08.922].
Viab ility of tissue s was asse sse d b y using the MTT
assay.
La p ré p aration-te st [08.922] a é té p ré p aré au
lab oratoire p ar mé lang e d e 3 g d e p oud re
[08.922] ave c 9 g d ’e au d istillé e . 5.35 µl d e la
préparation-te st ont é té ap p liq ué s à la surface d e s
épidermes.
Traitement “48h - 2 applications” : application du
p rod uit à J0 e t J1. Incub ation d e s tissus à 37°C
(48h). Test MTT à J+2.
The te st-form ulation was p re p are d b y m ixing 3g of
te st p owd e r [08.922] with 9 g of d istille d wate r.
5.35µl of the te st-formulation we re ap p lie d d ire ctly
to the surface of the stratum corneum.

Une trè s lé g è re d iminution d e la viab ilité d e s
é p id e rme s a é té e nre g istré e ap rè s traite me nt d e s
tissus ave c le p rod uit [08.922] (Fig 1). La ré p onse
MTT est réduite de 17%, après 48h de contact.

(Fig . 1). The tissue MTT re sp onse was re d uce d b y
17% after 48h exposure.

3.2 ACTIVITE DEPIGMENTANTE - DEPIGMENTING EFFECT OF TEST PRODUCT

SKINETHIC™ (lot n° 09 022M 0108 , surface 0.5 cm2)
répartis en 3 lots (3 épidermes/lot) :
- Lot [Témoin] : épidermes non traités
- Lot [08.922] : é p id e rme s traité s ave c [MASQUE
PEEL-OFF ECLAIRCISSANT]
- Lot [Contrôle Positif] : é p id e rme s traité s ave c
MELANEX® TRIO Crème.
The assay was p e rform e d on SKINETHIC™ RHTEps
(Batch n° 09 022M 0108, surface 0.5 cm 2) d ivid e d into
3 experimental groups (3 tissues/group) :
- [Control] group : untreated epidermis.
- [08.922] Tre ate d g roup : e p id e rm is tre ate d with
[MASQUE PEEL-OFF ECLAIRCISSANT]
- [Positive Control] g roup : e p id e rm is tre ate d
with MELANEX® TRIO cream.
Le s p rod uits te sté s ont é té ap p liq ué s, te ls q ue ls, à
raison d e 5mg / cm 2, sur le stratum corne um d e s
épidermes. Les formules-tests ont été appliquées à
J0, J1, J2, J3, J6 e t J7. Le s p aramè tre s analysé s
(coloration d e s é p id e rme s, taux d e mé lanine ) ont
été mesurés à J8 (après 6 applications).

Le traite me nt d e s tissus p ar la p ré p aration
[MELANEX TRIO] se trad uit p ar une imp ortante
d iminution
du
taux
de
mé lanine .
Comp arative me nt aux é p id e rme s té moins, le taux
d e mé lanine e st d iminué d e 55% ( p 0.01, te st t d e
Student). Ce résultat confirme la bonne réactivité
des tissus mis en œuvre dans l’essai.

[MELANEX TRIO] le d to a sig nificant d e cre ase in
m e lanin conte nt. Com p are d with control, m e lanin
le ve l was d e cre ase d b y 55% afte r top ical
application (Stud e nt’s t te st, p 0.01). This re sult
confirm s the g ood re activity of the tissue s all ove r
the experiment duration.
EPIDERMES TRAITES AVEC [08.922]:
La formule [08.922] manife ste une ne tte activité
d é p ig me ntante p uisq ue le traite me nt d e s
é p id e rme s se trad uit p ar une d iminution d e 28%
d u taux d e mé lanine . Ce t e ffe t d u p rod uit [08.922]
